Is there a way to set the plugin to display a single image in the gallery?

For Example:
{gallery}folder/image.jpg:::1{/gallery}

I will have a master gallery at the top of the page, but then as the article calls out specific functions, that particular image will be displayed.

I believe that you need someting like SIG Pro. Take a look here:

The question is how do I load up ONE image in a folder?

Or simply "point" to a specific image in a gallery as the thumb?

You can rename your photos like photo1.jpg, photo2.jpg photo3.jpg etc. If you select {gallery}folder/yourfolder:::1{/gallery} it will always shows on the frontend the photo1.jpg. This way rename photo1 your selected photo so as to be "pointed" on your page.
